L'intestin constitue la frontière entre le monde extérieur et notre organisme. Sa fonction première est d'assurer le transport des nutriments aux différents organes afin qu'ils en tirent l'énergie nécessaire à leur fonctionnement. En moyenne, les lipides (ou graisses) contiennent 2,25 fois plus d'énergie que les sucres et des processus complexes permettent leur cheminement de la lumière intestinale à la circulation du sang pour finalement rejoindre les organes. C'est ainsi que suite à l'ingestion des aliments lipidiques, on assiste à leur digestion en nutriments extrêmement simples, puis à leur assimilation dans les cellules intestinales pour être pris en charge par leurs véhicules naturels et essentiels, les chylomicrons. Globalement, l'absorption intestinale des lipides est un processus efficace. Néanmoins, notre laboratoire a bien démontré que des aberrations intracellulaires surviennent souvent : ils peuvent soit réduire dramatiquement le transit des lipides (ce qui engendre des malabsorptions sévères conduisant à la malnutrition), soit accélérer leur passage, augmentant de la sorte les taux circulants et causer des désordres lipidiques (dyslipidémies) postprandiaux. C'est le cas de l'obésité, la résistance à l'insuline et le diabète du type 2. Récemment, il s'est avéré que l'appareil gastrointestinal n'est pas un système inerte; ie, il est capable de synthétiser une multitude d'hormones susceptibles d'influencer la satiété, la prise alimentaire, la vidange gastrique, l'acidité et plusieurs évènements digestifs. Étonnement, l'impact des hormones gastrointestinales (GLP-1, GLP-2, PYY et ghréline) sur les processus intracellulaires gouvernant la formation des chylomicrons n'a pas été à ce jour explorée. C'est précisément le défi que relève le présent programme de recherche, dans l'espoir de trouver de nouvelles cibles thérapeutiques contre l'obésité, un problème majeur de santé publique. Nos recherches définiront le rôle des hormones gastrointestinales dans le contrôle des différents évènements prenant place dans les cellules absorptives de l'intestin menant au transport des graisses.
