本研究の目的は皮膚科領域の各種腫瘍に於いて癌抑制遺伝子のへテロ接合性の消失と遺伝子変異を検索することにある。様々な症例を出来るだけ数多く検索するためにホルマリン固定パラフィン切片よりDNAを抽出する方法を採用した。昨年度、DNAを確実に抽出する方法の条件設定がすみ、皮膚腫瘍発生に関連している可能性のある癌抑制遺伝子の検索を行った結果,様々な腺癌の初期に関係していることが解明されつつあるが皮膚科領域では未だ研究されていないAPC遺伝子をその検索対象とすることにした。転移性皮膚腫瘍,外陰パジェト病,汗管系悪性・良性腫瘍,悪性黒色腫,有棘細胞癌のそれぞれの対象にする標本の切片を実体顕微鏡下で確認しながら正常部と腫瘍部を分離して核をかきとり、プロテナーゼK処理によりDNAを抽出し、PCRによって増幅したDNAを制限酵素処理することによりAPC遺伝子のへテロ接合性の消失の検索を行った。検索を行った全57例のうちへテロ接合性について検討可能であったのは、32例であり、その内APC遺伝子が関与している可能性があったのは汗管系悪性・良性腫瘍 5例と転移性腫瘍のみであった。この5例について制限酵素処理する前のPCR産物をSSCP用の機器を用いて、PCRによって増幅された範囲に遺伝子配列の突然変異がみられるかどうかの検索を行ったが、様々な条件設定を試みてみたがSSCPでは、ヘテロ接合性消失が認められた症例と認められなかった症例の間で、泳動結果に差を見いだすことはできなかった。
