We request DFG funding for a Molecubes small animal PET/CT (ß- und X-Cube combination, "benchtop PET CT integrated platform"), a small labspace molecular imaging device with each one component for PET and CT.Our research is focused on translational theranostics, based on small radioligands for diagnosis and treatment of cancer. Translational research aims at: Prostate cancer is one of the leading causes of cancer-related death among men worldwide. We developed a mouse model of prostate cancer with high expression levels of PSMA. Small animal PET/CT will be used to assess alternative routes of tracer application in mice. We further aim to evaluate improved efficacy through radioligand drug combination therapy with VE-822 and Enzalutamide. Fibroblast activation protein (FAP) is expressed at high levels in fibroblast-associated malignancies. Chinolin-based radiolabeled inhibitors of FAP (FAPI) can be applied to accurately assess extent of breast cancer, pancreatic cancer, or ENT tumors using positron-emission-tomography (PET). We will develop a mouse model of human cancer to test various radiolabeled FAP-inhibitors for their biodistribution and accuracy. In collaboration with other University institutions, we will use small animal PET/CT imaging to assess immunstimulatory calciumphosphate nanoparticles, avatar-models of lung cancer, murine melanoma models, and off-line dose assessments for proton beam therapy.
