项目摘要

Rats were inoculated 2 weeks (three times per week) with Porphyromonas gingivalis (P.g.) that is thought as the periodontopathic bacteia, after dental calculus had been formed. Before inoculation, the specific pathogen were reduced with sulfamethoxazole/trimethoprim and were eliminated oral pathogenic bacteria. After the medicament of anti-bacterial drugs, P.g. was inoculated. The dental calculus formation food by the prescriptions of Bringer et al (1971) was given to rats for 4 weeks. Rats were divided into 4 groups, 1 group was negative control (bacteria was not inoculated), 3 remaining groups were changed breed periods. P.g. was cultivated on supplemented blood agar (Colombia CNA based with 5.0μg/ml hemin, 0.5μg/ml menadione and 5% defibrinated sheep's blood). For experiments, bacteria were grown on supplemented blood agar at 37℃ in an anaerobic chamber under an atmosphere of 10% carbon dioxide, 10% hydrogen and 80% nitrogen. The rats were inoculated live P.g.suspension in 5% carboxymethyl-cellulose in PBS solutions. The bacteria were inoculated directly in the oral cavity of rats. It was estimated the adhesion of calculus with the method of Bringer et al(1971), and evaluated the absorption degree of an alveolar bone defects. Although the adhesion of the calculus is adhering a lot on the side of the lower jaw as a result, the settlement of the P.g.bacteria was in a decrease trend gradually. Also, the adhesion degree of the calculus was in a big trend, without being related to low, in the case that the absorption degree of the alveolar bone inoculated the P.g.bacteria.
将大鼠接种牙龈卟啉单胞菌(P.g.)2周(每周3次),即牙结石形成后的牙周致病菌群。接种前用复方新诺明/甲氧苄啶对特定病原菌进行灭活,并对口腔致病菌进行杀灭。抗菌药物的药物后,P.g.接种了疫苗将Bringer等(1971)处方的牙石形成食物给予大鼠4周。将大鼠分为4组,1组为阴性对照组(不接种细菌),其余3组为换种期组。P.G.在补充的血琼脂(基于5.0μg/ml氯化血红素、0.5μg/ml甲萘醌和5%脱乙酰化羊血的哥伦比亚CNA)上培养。对于实验,在37℃下,在10%二氧化碳、10%氢气和80%氮气气氛下的厌氧室中,将细菌生长在补充的血琼脂上。大鼠接种含5%羧甲基纤维素的PBS溶液中的活P.g.混悬液。细菌直接接种于大鼠口腔。采用Bringer等(1971)的方法评价牙结石的粘附情况,并评价牙槽骨缺损的吸收程度。虽然牙石的附着在下颌一侧的附着量很大,但P.g.菌的沉降量呈逐渐减少的趋势。在牙槽骨吸收程度方面,接种P.g.菌后,牙结石的粘附程度有大的趋势,与低无相关性。
