X-ray crystal structure of the complex between the Ca^<2+>-binding protein ALG-2 and the binding peptide of its interacting protein Alix was resolved, and the molecular basis of the Ca^<2+>-dependent binding was elucidated. The ALG-2 dimer has one binding site in its monomer molecule, and was found to function as an adaptor to bridge Alix and TSG101 in the presence of Ca^<2+>. It was also found that ALG-2 bridges Sec31A and annexin A11.
