This project will explore a new method of obtaining 3D Fluorescence correlation Spectroscopy (FCS) measurements utilising a new STED-FCS-FLIM microscope, which, when compared against typical methods, will allow for much quicker measurements whilst retaining high spatial resolution.Increasing the rate at which these measurements are obtained is essential as the current, slower, methods are not quick enough to capture and quantify many of the essential molecular processes and dynamics which occur within the cell membrane in the required detail. One such process which occurs within the cell membrane is insulin-regulated membrane trafficking, which is known to be impaired with diseases such as Type 2 diabetes, where insulin transfer is somehow inhibited. This project aims to increase our understanding of such processes within fat cells, seeking to therefore increase our understanding of the mechanisms behind diseases such as Type 2 diabetes, through an increased understanding of molecular dynamics within cell membranes, and the development of new methods for 3D super-resolved measurements.
