This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Brief Background: In this study we will investigate if the accumulated fat in muscle and liver in people who already have a reduced effect of insulin (an important hormone for regulation of glucose and fat processing in the body), can be modified by intake of specific nutrients, e.g. fish oil and amino acids (building blocks of protein). Specific Aims and Procedures: The main goals of this project are to determine 1) if supplementation of protein and fish oil will improve the effect of insulin in the body and decrease muscle and liver fat accumulation in subjects with reduced sensitivity for insulin, and 2) if supplementation of amino acids and fish oil will improve the effect of insulin in the body and decrease muscle and liver fat accumulation in subjects with reduced sensitivity for insulin. Experimental Design. The participant will take a nutrient supplementation each day for 16 weeks. The supplementation will be given as pills and/or drinks and will consist of either: 1) fish protein, 2) amino acids, 3) fish protein + fish oil, or 4) amino acids + fish oil. The following day different tests will be performed. The tests include measurement of body composition by a body x-ray scan (DEXA), and test of the sensitivity for insulin measured by an oral glucose tolerance test (OGTT). Significance: The study will help to better understand the causes of diabetes and how diabetes can be prevented.
