INSULIN RESISTANCE, AGING AND THEIR RELATION TO MUSCLE AND LIVER LIPID CONTENT

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The development of an age-related insulin resistance is a well-documented finding in the literature. Further, there is a relationship between the degree of insulin resistance and the extent of intracellular lipid accumulation in the muscle and liver. We, therefore, propose to determine the relationship between insulin resistance and aging as a function of muscle and liver lipid contents in apparently healthy individuals. For this purpose we will study a group of young (18 to 35 years of age) and a group of old (over 65 years of age) individuals. The extent of insulin resistance will be determined on the basis of procedures involving the oral glucose tolerance test, while muscle and liver lipid contents will be determined using proton magnetic resonance spectroscopy. We also plan to study a group of elderly individuals with type 2 diabetes mellitus who will undergo the same testing procedures as the healthy individuals. Data collected from the latter group will allow testing the hypothesis that there are common underlying factors related to muscle and liver lipid contents between aging and type 2 diabetes mellitus.
