FLUORESCENCE MICROSCOPY OF PROTEINS INVOLVED IN GOLGI-TO-VACUOLE VESICLE TRAFFIC

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We are interested in the role of known vacuole fusion factors within the AP-3 vesicle trafficking pathway. While we have identified in vitro interactions between the AP-3 vesicle coat complex and a subset of fusion factors, whether these interactions occur in vivo remains unknown. Our goal is to use fluorescence microscopy to determine the colocalization of AP-3 with these fusion factors in vivo and at which cellular compartment this may take place. This information will provide insight into whether AP-3 interacts with the fusion factors in vivo and the possible function of these interactions.
