This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The JEOL3000SFF electron cryomicroscope is a liquid helium microscope opearted at 300 kV and specimen temperature of 4.2K. This instrument is presently dedicated to high resolution single particle image data collection. We have 2 well-trained scientists using this instrument routinely. The instrument has been in high performance mode throughout most of the year. The downtimes have attributable mostly to the dirty objective aperture, camera jam and malfunction of the automated specimen cartiledge loading device. The biolgoical projects include icosahedral virus particles and chaperonin particles which are aimed at 4 ¿ resolution data.
