This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The overall aim of this study is to examine the gentic and environmental influences underlying the human behavioral traits of aggression and impulsivity in a population-based sample of twin subjects (PennTwins: a cohort developed by the PI). In this study, both aggression and impulsivity will be examined using self-report (life history and personality inventory) and laboratory measures of aggression and impulsivity; measures of 5-HT will include measures related to the 5-HT Transporter. In Phase I of this project, twins will be administered a variety of questionnaires realted to impulsivity and aggression and other related behavioral variables. In Phase II, 420 twin (140 same sexed MZ, 140 same sexed DZ, and 140 opposite sexed DZ if gender differences in genetic influences are detected in Phase I or 210 same sexed Mzand 210 same sexed DZ if no gender differences in genetic influence are detected in Phase I) pairs will be studied in our Clinical Neuroscience Research Unit at The University of Chicago. Data will be collected from various experimental procedures, including laboratory analog assessments of aggression (Point Subtraction Aggression Paradigm: PSAP, Taylor Aggression Paradign: TAP, and Self Aggression Paradigm: SAP) and impulsivity (Immediate Memory Task: IMT, Go-Stop Task: GST, and the Single Key Impulsivity Paradigm: SKIP) Platelet 5-HT Transporter structure (i.e., number of 5-HTT binding sites: Bmax), function (i.e., velocity of 3H-5-HT Uptake: Vmax), and genotype (5-HTT Promoeter DNA Polymorphism) will be obtained to determin whether measures of serotonergic function demonstrate evidence of heritability and to examine the nature of a genetice influence on the inverse phenotypic correlation between 5-HT and aggression/impulsivity previously established over two decades of previous research. CRC resources are requested primarily for ancillary services in obtaining blood samples for Platelet 5-HT Transporter Binding/Uptake Velocity and in obtaining blood samples for Platelet 5-HT Transporter Binding/Uptake Velocity and in obtaining blood samples for DNA Extraction for confirmation zygosity status and for genotyping of 5-HT Transporter polymorphisms.
