This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The purpose is to test various priming and boosting regimes in the DNA prime-VSV boosting systems in candidate AIDS vaccines. In June 2007, 30 animals were obtained with the intention of comparing boosting efficiency of VSV-Gstem, a non-replicating vector and replicating-VSV vectors in animals that were primed with DNA delivered via electroporation. Heterologous DNA/VSV immunizations were also compared with the homologous DNA. The animals were divided into 5 groups of 6 each. Groups 1, 2, & 3 received 3 DNA priming immunizations every 30 days. Groups 4 & 5 did not receive a priming immunization. At 5 month post first vaccination, groups 1 & 4 received a VSV-Gstem boosts, groups 2 & 5 received a replicating-VSV boost, and group 3 received a DNA boost. Group 3 received 2 additional boosts in February 2008, while the other 4 groups will receive boosts in March 2008. Blood samples have been collected in order to monitor the immune responses of these animals. The cell-mediated immune responses will be followed through July 2008. This program is being terminated by Wyeth Vaccines. The intention is to move the research to another vaccine company.
