This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Mycobacteria vectors offer considerable promise as a mucosal vaccine, but the utility of these vectors has to date been limited because of poor antigen expression, vector instability, and the prevalence of pre-existing Anti-BCG immunity. Mycobacteria have features that make them attractive as potential HIV-1 vaccine vectors. Most importantly, they have been used successfully as vaccines. The attenuated, non-pathogenic Mycobacterium bovis BCG has been widely used as a vaccine for tuberculosis (TB) and leprosy. It can also be readily engineered to stably express transgenes.
