In the last decades, biomaterials have been successful to improve quality of the life for millions. However, very rare are the applications for which biomaterials and medical devices are completely successful. Failures or clinical complications are often observed, with different incidences related to the applications. This discovery program is very solidly based on advancing material science and engineering aspects applied to health, and in particular to the medical field. The proposed long-term vision is to deepen and consolidate a new 3 blocks-research program for: 1) investigating the parameters allowing the development of a highly-adherent and strongly-cohesive coating mainly based on plasma-based surface modification for metallic devices; 2) designing a new class of bioabsorbable metals specific for temporary devices; and 3) developing new strategies for testing, assessing and predicting the biological performances of the newly developed coatings and metals when implanted in humans. The overall goal of blocks 1 and 2 is to provide, at short term (1 to 5 years), new coatings and new materials to improve the clinical performances of medical devices. The overall goal of block 3 is to generate at medium term (5 to 10 years) advanced (human) cell-based in vitro alternative to (animal) in vivo tests. This research is extremely original and innovative and is likely to lead the ground-breaking advances in assessing the fate and impacts of newly-developed coatings and materials in a tissue-regenerated physio- and patho-logical 3D cultured vascular wall models from human adult cells. The original scientific knowledge generated is sure to improve risk assessments and support the development of regulatory guidelines by government agencies. Moreover, the proposed research program is planned to result in a polyvalent multi-facetted (coatings-materials-tests) platform susceptible to be applied to other fields and applications, where specifically-designed materials and coatings with targeted properties are required. In this proposal, we both advance knowledge and train high quality personnel in key areas of strength for Canada. Our research environment is exceptional and particularly adapted to meet the new Global market and challenges we are facing today. More than 100 HQP were trained since 2015: Today, they pursue positions in academia, industry and government by using daily advanced knowledge, leadership, fast adaptive skills, global network connections and strategic thinking for which they have been trained with us. In conclusion, our Laboratory is recognized to impact the clinical and industrial practice in reparative surgery, and to open new doors in Biomaterials Science, patent new inventions and propose licenses, welcome established and young scientists with invited lectures at international conferences, closely collaborate with world leaders in academia and industry, and regularly assist in providing advices to regulatory governmental agencies.
