In this research program, phytochelatins, glutathione and related thiol-peptides involved in pollutants cellular sequestration and detoxification mechanisms will be investigated with ultra-high-performance liquid chromatography. This research instrument is composed of a core system permitting high-efficient separation of these proteins through specialized columns, an automated sample injection for high-throughput analysis, and a fluorescence detector providing high-sensitive qualitative and quantitative detection of these proteins. This analytical approach will be used on green algae strains exposed to different concentrations of aquatic pollutants to identify the different biochemical properties of algae related to the transport and sequestration of aquatic pollutants in the cell vacuole. The results obtained by this approach will be used to develop a fundamental model which identifies the best algal strain characteristics to obtain the most efficient vacuole sequestration of pollutants. This model will be used to develop new biotechnological approaches to remediate contaminated wastewater. This environmental biotechnology using algal detoxification mechanisms can be applied to prevent the pollution of aquatic ecosystems and protect the quality of the environment.
