项目摘要

Lactobacillus gasseri is known to be exist in human intestine, so the strain is widely used for the construction of functional yogurt as probiotics lactic acid bacteria (LAB) in Japan. In this study, it has been clarified the existence of three type of lactose hydrolyzing enzymes beta-galactosidae (β gal), phospho-beta-galactosidase (P β gal) and phospho-beta-glucosidase (P β glc) in LAB including L.gasseri. In this research, PbgA (phospho-beta-galactosidase A) was first isolated from L.gasseri ATCC33233^T and investigated by kinetics analysis. As the expression rate was very low for PbgB in the L.gasseri ATCC33233^T, we prepared the enzyme by expression with Lc.lactis subsp. cremoris and analyzed K_m and V_<max> values.We also investigated the adhesion activity of LAB to human colon mucin by a biocencer BIACORE which is one of most important requirement of probiotics. We could selected many adhesive LAB with high affinity to human mucin with ABO blood type A,B and O type.In the last stage of our research, to prevent the lactose intolerance in Japanese, we did mass-screening of LAB with high lactase activity from more than 200 strains of LAB isolated human intestine. Finally, we selected Lb. mucosae OLL2848,Lb. gasseri OLL2836 and Lb.gasseri OLL 2948 strains and constructed the new type of functional yogurt by using these three LAB strains.It was expected the decrease of symptoms of lactose intolerance especially to elderly peoples.
加氏乳杆菌(Lactobacillusgasseri)是一种存在于人体肠道中的益生菌,在日本被广泛用于功能性酸奶的构建。本研究阐明了包括加氏乳杆菌在内的乳酸菌中存在β-半乳糖苷酶(β gal)、磷酸-β-半乳糖苷酶(P β gal)和磷酸-β-葡萄糖苷酶(P β glc)三种乳糖水解酶。本研究首次从加氏乳杆菌ATCC 33233 ^T中分离得到PbgA(phospho-beta-galactosidase A,磷酸-β-半乳糖苷酶A),并对其进行动力学分析。由于PbgB在加氏乳杆菌ATCC 33233 Δ T中的表达率非常低,我们通过用乳酸乳杆菌乳酸亚种表达来制备该酶。并<max>利用BIACORE生物活性测定仪研究了乳酸菌对人结肠粘蛋白的粘附活性。本研究的最后一个阶段,为了预防日本人乳糖不耐受,我们从200多株人肠道分离的乳酸菌中大量筛选了具有高乳糖酶活性的乳酸菌。最后,我们选择了LB。mucosae OLL2848,Lb.利用乳酸菌OLL 2836和Lb.gasseri OLL 2948构建了一种新型的功能性酸奶,有望减轻乳糖不耐症,尤其是老年人的乳糖不耐症。
