Recently the target proteins of mitochondrial sirtuins (SIRT3,4,5) were clarified respectively, still a lot of things are the mysteries for the function of mitochondrial sirtuins. To investigate the role of the mitochondrial sirtuins, we have comprehensively disrupted the gene of mitochondrial sirtuins using DT40 cells. We succeeded in single sirtuin-deficient cells and double sirtuin-deficient cells by present. The little delay was seen in growth in double sirtuin-deficient cells compared with wild type cells though these deficient cells were able to grow everything.
