This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. During systemic hypoxia, sympathetic nerve activity is increased accompanied by increases in skeletal muscle interstitial adenosine and norepinephrine. It is not known whether the increases in interstitial adenosine come from vascular endothelium, from skeletal muscle fiber or from extra cellular conversion of ATP. In this study, we propose that through simultaneous measurement of norepinephrine and adenine nucleotides (ATP, ADP, and AMP) from intramuscular microdialysis probes during three distinctly different sypatho excitatory interventions we will obtain the necessary to differentiate unique vascular responses to hypoxia. The three different 15 minute stimuli that will be used include: 1) systemic hypoxia; 2) intravenous infusion of sodium nitroprusside; and 3) lower body negative pressure. 16 volunteers will be studied.
