Project Summary The goal of the COBRE-II parent award at MDIBL is to study the intersection of stress, aging, and regeneration using comparative functional genomic studies of appropriate model organism systems. In the current proposal, Dr. Murawala (RPL within COBRE II at MDIBL) will team up with Dr. Arturo Hernandez (Faculty scientist III, MHIR) and Dr. Joel Graber (Director, MDIBL Computational Biology Core) to perform comparative studies the effect of thyroid hormone-induced metamorphosis in axolotl and mouse using next-generation methodologies. Dr. Murawala will focus on understanding the thyroid-hormone induced changes in axolotl skin using multiome and spatial transcriptome technology of 10X Genomics, Dr. Hernandez will perform complementary experiments in mouse models, and Dr. Graber will refine the axolotl-mouse orthology mapping and will use this updated information to analyze and compare multiome and spatial data from the Murawala lab and Hernandez labs. The work proposed here will allow us to generate valuable comparative datasets that will 1) provide crucial insights into pathophysiology of skin upon abnormal thyroid hormone signaling and 2) form the basis for future larger grants aimed at elucidating the role and underlying mechanisms by which thyroid hormones regulate tissue maturation and regenerative capacity.
