Nuclear receptors are part of a family of ligand activated proteins that reside in the nucleus of cells and control the expression of specific genes involved in diverse aspects of metabolic functions. To achieve such control, nuclear receptors are directly involved in making contact with target DNA in order to activate or repress specific sets of genes in cells. As such, investigation of how key genes controlling various steps of metabolic pathways are regulated through nuclear receptor activations will greatly improve our understanding of gene transcription and dynamics.***In the laboratory, we have been studying in particular the two estrogen receptors (ERa, ERb) and the PPAR family members based on their respective potential to modulate response to hormones and to control energy metabolism. The proposed equipment is essential to investigate the functional aspects and specific modifications of nuclear receptors in the context of gene transcription, complex assembly, mitochondrial activity and cell phenotypes. The equipment will also allow specific metabolic measurements in respect to other research projects undertaken by the co-applicants. Therefore, several cell biology approaches will be supported in several laboratories to address the role and impact of key effectors and conditions in every research project. The formation and training of highly qualified personnel will also be favorized with the use of the proposed equipment.***Our long term objective is to improve our understanding of how upcoming signals impact cellular processes, in order to provide novel insights in the control of gene network and metabolic outcomes.
