(a) The iIACS machine has been improved, which far surpasses the previous state-of-the-art iIACS machine in system performance, expanding the range of applications and discoveries. Its throughput and sensitivity are 20 times superior to those achieved in previous system.(b) By employing the VIFFI microscope, the iIACS machine has higher imaging performance, holds promise for diverse applications in immunology, microbiology, stem cell biology, cancer biology, pathology, and synthetic biology. The imaging performance of the iIACS machine was validated using various cell types, like the FISH images of the Jurkat cells, imaging and sorting for the budding yeast cells.(c) I have developed a viscoelastic fluid-based 3D focuser, and employed it for single-particle Raman analysis, as well as into the flow module in VIFFI platform, which can enable high-throughput single-cell imaging and analysis with higher stability.
