1.Polyclonal antibodies against rat 5 alpha-reductase type 1 were generated in rabbits. Using the antibodies, the localization of 5 alpha-reductase was immunohistochemically studied in the rat tissues. The results are as follows.(1) In the anterior pituiary gland, the positive immunoreaction was present in gonadotrophs. Castration brought about the increased staining intensity.(2) In the ovary, the enzyme was present in most theca interna cells and some of granulosa cells.(3) In the prostate, the epithelial cells of ventral and dorsal gland were positively stained but those of lateral ones were negative. After castration, the number of the positively stained cell decreased, but the intensity of the staining apparently increased. Electron microscopic immunocytochemistry revealed that the enzyme was present in the membranes of endoplasmic reticulum.(4) In the testis, the enzyme was localized in Leydig cells.(5) In the nervous system was mostly present in Schwann cells and oligodendrocytes.2.Electron microscopic immunocytochemistry demonstrated the colocalization of ATP synthase and cytochrome P-450 of side chain cleavage enzyme in the mitochondria of rat and bovine adrenocortical cells.3.Prostaglandin endoperoxide synthase was immunolocalized in mast cells, smooth muscle cells of muscularis mucosae and inner muscle layr, endothelial cells of arteries, nerve cells of myenteric plexus and perioneal epithelial cells of bovine intestines.
