We studied the effect of acetoaldehyde (AcCHO) on gene expression of matrix metalloproteinase-2 and -9(MMP-2 and -9) and tissue inhibitors of metalloproteinase-1, -2 and -3(TIMP-1, -2 and -3) in cultured Ito-cells isolated from surgically resected human liver. Cultured Ito-cells expressed mRNA of MMP-2, TIMP-1 and TIMP-3 respectively. AcCHO did not show any stimulating effect of type IV collagen synthesis, whereas inhibited the mRNA levels of MMP-2, TIMP-1 and -3. The gene expression of MMP-9 and TIMP-2 could not detect in cultured human Ito-cells. Decreased levels TIMP-1 and -3 may be in consequence of decline of MMP-2. These data suggest that suppressed gene expression of MMP-2 may contribute to enhance the type IV collagen deposition in alcoholic liver fibrosis.
