This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Specific Aims - The growing understanding of replication complexes presented in last year¿s progress report posed important questions and opportunities that cannot be addressed by conventional two dimensional (2D) EM imaging. All positive-strand RNA [(+)RNA] viruses replicate their genome in association with host intracellular membranes that are rearranged into discreet structures. The Nodavirus Flock House virus (FHV) induces the rearrangement of mitochondrial outer membranes into spherule structures during infection of insect cells. RNA replication occurs at the outer mitochondrial membranes in these FHV infected cells. Our aim is to use electron microscope tomography to better understand the mechanism of FHV RNA replication.
