This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
It is generally accepted that high-grade prostatic intraepithelial neoplasia (HGPIN) is a precancerous lesion. Several groups, including ours, have demonstrated that the majority of HGPIN lesions have reduced or absent levels of annexins (ANX) I and II proteins. However, a causative link between the reduced expression of annexins and HGPIN has not been established. To investigate this possibility we have silenced ANX I or II expression in benign prostatic epithelial cells and examined the glandular architecture and cellular polarity in 3-dimentional (3-D) culture. To determine if silencing ANX I and II expression in normal prostatic epithelial cells alters development of acinar structures in 3-D culture, we will investigate 3-D structures by using confocal laser scanning microscopy (LSM), multiphoton microscopy (MPM) and MPM/optical coherence tomography (OCT) system.
Preliminary data obtained with LAMMP technologies were used for NIH grant submission.
