This subproject is one of many research subprojects utilizing the

Backgrounds: The efficiency of controlled propagation to produce rhesus monkeys of particular genotypes can be maximized by use of cryopreserved spermatozoa collected from specific males to inseminate appropriate females. This study was designed to assess the effects of various cryopreservation protocols on the post-thaw survival of rhesus macaque semen. Methods: Multiple ejaculates of five mature males were collected and frozen by each of four different methods. Semen was stored in liquid nitrogen and then thawed. Total and progressive motility were assessed immediately after thawing, after 30 min, 60 min, 240 min and again 18 hours later.
Results: Cryopreservation significantly reduced motility in all samples regardless of the cryopreservation method. Conclusion: The response of different ejaculates of the same male to each method proved to be consistent. However, the method of freezing was a significant factor in determining the immediate motility, although this difference had disappeared after four hours when motility was nearly the same in all samples. Moreover there was a significant interaction between individual males and freezing method suggesting that developing a single freezing protocol that is universally suitable may be difficult.
