During this period, the project team has completed optimization of experimental conditions of a high-throughput screening platform capable of measuring dynamic states of chromatin accessibility in cells using an enzyme-based approach (Assay for Transpose-Accessible Chromatin (ATAC)). The group has validated a crude preparation approach for the ATAC DNA that saves significant cost and increases throughput. Using the optimized assay, a pilot screen was performed using an epigenetic-focused small molecule library, and active compounds are undergoing further characterization.
