We have been investigating on correlation between radiation sensitivity and various cell cycle relating and or apoptosis relating proteins including p53, p21, BAX BCL2. Although some correlation have been demonstrated, The correlation was nor so significant than we expected. Hence another cell cycle modulating pathway including TGFβ, p27 SMAD was investigated with imunohistochemical methods.Methods. A total of 202 biopsy specimens obtained from 77 patients with squamous cell carcinoma of the cervix before and during radiotherapy (RT) were investigated for the expression of p21, p27, and p53, using immunohistochemical methods. Results. Cancer cells, and degenerated or swollen cancer cells caused by RT that were positive for p21, p27, and p53 showed an intranuclear reactivity. Degenerated or swollen cancer cells at 27Gy showed stronger p53-positivity than cancer cells before RT. The mean p27-labeling index was significantly decreased at 27Gy, conversely, the mean p53-labeling index was s … More ignificantly increased at 27Gy. The high p27-labeling index before RT was significantly associated with good disease free and metastasis free survivals. The high p53-labeling index before RT was associated with a poor overall survival Conclusions. The high p27 and low p53 expression in cancer cells before RT are regarded as a preferable predictive factor for the prognosis of patients with cervical squamous cell carcinoma treated with RT alone. The mean p27 and p53 indexes change in reversal fashion during the periods from initiation of RT to at 27Gy.A monoclonal antibody, MIB1, recognizes Kt-67 antigen in routinely processed paraffin sections after microwave treatment. The MIB1 labeling index represents the growth fraction of the cell population. Methods. A total of 150 cervical biopsy specimens taken from 75 consecutive patients with cervical squamous cell carcinomas before radiotherapy (RT) and after 9Gy of RT were investigated. Results. & Conclusions. The high growth fraction after 9Gy of RT is regarded as predictive factors for good prognosis of patients with cervical squamous cell carcinoma patients with RT alone. Less
