REDUCING ANNEXIN I OR II IN 3-D CULTURE BENIGN PROSTATIC EPITHELIAL CELLS

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. It is generally accepted that high-grade prostatic intraepithelial neoplasia (HGPIN) is a precancerous lesion. Several groups, including ours, have demonstrated that the majority of HGPIN lesions have reduced or absent levels of annexins (ANX) I and II proteins. However, a causative link between the reduced expression of annexins and HGPIN has not been established. To investigate this possibility we have silenced ANX I or II expression in benign prostatic epithelial cells and examined the glandular architecture and cellular polarity in 3-dimentional (3-D) culture. To determine if silencing ANX I and II expression in normal prostatic epithelial cells alters development of acinar structures in 3-D culture, we will investigate 3-D structures by using confocal laser scanning microscopy (LSM), multiphoton microscopy (MPM) and MPM/optical coherence tomography (OCT) system
