This application requests the purchase of an ultra-low temperature freezer to maintain the long term viability of bacterial cultures used in research supported by NSERC, industry and industry-associations. The research, in which resuscitated cultures are used, is focused on the microbial ecology of foods where both pathogenic and spoilage organisms are studied to enable the safe extension of the shelf-life of food products. Work previously conducted and that planned examine factors in the food animal environment that influence food safety. The current equipment request is motivated by the age and unreliability of the freezer presently used, as well as its overall importance in providing safe storage for a large number of bacterial isolates of known industrial value (fermentations) as well as those with known biochemical or virulence characters. Reference strains from accredited international collections are also housed in this storage. Dependable storage of cultures that are readily accessible provides back-up in case of loss, and allows for continuity in the progress of research, particularly where environmental isolates are obtained from different sectors of the food chain. This resource permits comparisons of the origins of organisms and allows for examinations of antimicrobial susceptibility that can lead to better means for bacterial control.
